Patented computer-driven pump with sensor data features a compact design for enhanced replicability in eDNA sampling. To equip citizen scientists and commercial vessel operators with an easy-to-use eDNA sampling solution, we developed the semi-automated DNAutic device. Designed for seamless operation in freshwater and marine environments, it is fully compatible with Smith-Root self-preserving eDNA filter packs.

eQuip sampling system & filter kits

This original underwater sampling system was developed by Sequench with support from the Callaghan Innovation R&D fund. It allows concentrating waterborne eDNA without onboard pumps. Our patent-pending design uses a Venturi-based suction system: a narrow throat followed by up to three perimeter sampling openings. The pressure differential created by the Venturi effect enhances water intake and ensures consistent flow through the filter units mounted on each intake.

eQuip underwater sampler operates at cruising speeds up to 12 knots, delivering ~5–12 L/min per intake (speed-dependent) for high-volume eDNA capture and clean field workflows. Its teardrop hydrodynamic body minimises drag, while inverted planform lateral wings and directional fins keep the unit stable and on course during tow. The filter units are mounted onto outer parts of different sample collection systems with minimal handling interference, thus minimizing risk of contamination.

Proven on small motorboats, sailing yachts, and oceanographic vessels across lakes, coastal waters, and open ocean. With a weight depressor, the system can be submerged and towed to ~30 m depth.

The filter kit consists of individually packed universal filter membrane holders made of recycled biomass-based plastic (PLA), designed to accommodate standard 47 mm diameter filter membranes. The inner ring securely holds the membrane during sampling and can be easily detached from the base by flipping the lip, allowing the membrane to be isolated for downstream analysis without excessive handling. The lip on the base also serves as a handle, enabling straightforward insertion and retrieval of the filter from packaging and sampling devices.

For sampling, filter units are removed from the pouch, inserted into the sample mounts, and deployed at 6–12 knots for 5–10 minutes, depending on sampling requirements. Following retrieval, the filter unit is removed using the holding lip, excess water is shaken off, and the unit is returned to the desiccation pouch for storage until processing, or the membrane can be transferred directly into a tube containing preservation buffer. The storage bag should be securely sealed to ensure effective preservation. If freezing is the preferred method, the filter unit can be stored directly in the sealed bag at −20 °C.

For enhanced preservation, particularly when extended storage prior to laboratory processing is expected, or when targeting low-abundance or potentially degraded eDNA signals (e.g. fish or other vertebrates), a buffer spray option is available. This approach has been validated through extensive field trials. The buffer is supplied in a 10 mL spray bottle (sufficient for approximately 25 filters), with four sprays applied directly onto the membrane immediately after retrieval and prior to placement into the desiccation pouch.
